Description:

The maize event DP910521 was produced via site-specific integration to convey resistance to certain lepidopteran insect pests and tolerance to glufosinate herbicide.

To produce this event, in a first step, an insertion site sequence, the landing pad, was introduced via microprojectile bombardment. In a second step, the intended integration cassettes from plasmid PHP79620 were introduced by microprojectile bombardment as well. The insert comprises three expression cassettes, each featuring plant specific promotors and terminators:

• the cry1B.34 gene cassette (CS-cry1B1Ca19Db1-SYNTH) coding for an insecticidal delta endotoxin to mediate Lepidoptera resistance.

• the pat gene cassette (CS-pat-STRVR) coding for the phosphinothricin acetyltransferase to mediate tolerance to glufosinate herbicide.

• the pmi _ gene cassette (CS-pmi-ECOLX) coding for the phosphomannose isomerase as selection marker. The ubiZM1 promoter in the landing pad is utilized for the _pmi expression.

Conventional crosses and molecular characterization by SbS™ analysis confirmed stable insertion of a single copy of the intended DNA and the absence of additional unintended vector fragments in maize DP910521. Sanger sequencing was used to confirm the sequence.
