GE-MLO-KO Wheat was produced through transient expression of TALEN to create a knockout of the Mildew Resistance Locus (MLO) gene.
The expression constructs consisted of the TALEN expression cassette targeting sequences within the second exon of the Mildew Resistance Locus gene and the selectable marker expression cassette. Both cassettes contained the P-ubi1 promoter from maize and the T-nos terminator.
PCR analysis confirms that the construct was segregated away.
GE-MLO_KO Wheat is therefore a null-segregant that has improved disease resistance to powdery mildew.
