Rice B827 was produced by transforming the rice variety Tarom Molaii via particle bombardment with the two vectors pCIB4421 and pHygII. pCIB4421 contains a modified version of the insecticidal cry1Ab gene from Bacillus thuringiensis subsp. kurstaki. It is expressed under the control of the PEPC promoter from maize, leading to high-level expression in the leaves but not in the grain and confers resistant against lepidopteran pestes. The hpt gene, which was introduced with pHygII and is leading to a resistance to hygromycin, was used for selection after transformation.
Southern blot analyses indicate that the plants contain fewer than five, most likely only three copies of the cry1Ab gene, and that the majority of these copies are intact. There is no information on the copy number of the hpt gene in the plants. As both genes were transformed in separate constructs their copy number may vary.
