Rice event LLRice601 was produced by Agrobacterium-mediated transformation of rice variety Cocodrie with vector pGSV71 containing a single T-DNA.
The T-DNA construct contains the bar gene from Streptomyces hygroscopicus encoding the enzyme phosphinotricin acetyltransferase (PAT) conferring resistance to glufosinate herbicide.
Molecular characterisation of the genetic modification in LLRice601 demonstrates that a single copy of the intended T-DNA was integrated at a locus on chromosome 12 with a deletion of 19 bp in the rice genome.
Southern blot data suggests that the nos terminator (T-nos-RHIRD) is truncated, but sufficient to allow expression of the bar gene (CS-bar-STRHY). A second copy of the 35S promoter (P-35S-CaMV), or a fragment thereof, is inserted elsewhere in the genome. Southern blot analysis demonstrated the absence of any coding sequences from the vector DNA in the host genome.
