YAK729.6 is a genetically modified strain of Saccharomyces cerevisiae used for the large-scale production of a precursor of an analogue of human insulin.
YAK729.6 was developed through transformation of S. cerevisiae MT663 with the expression plasmid pAK729.6.16D. This plasmid contains a synthetic gene encoding a human insulin precursor and the triose phosphate isomerase (tpi) gene. This gene, inactivated in the recipient strain and restored in the YAK729.6 due to its presence on the plasmid, serves as selection marker.
The inserted sequences are only present in the plasmid, there is no integration in the chromosomal DNA; the entire plasmid is present in YAK729.6.
